Live nuclear Staining using Hoechst

Solutions Required

Stock solutions	Working solution
Hoechst 33342 Solution (20 mM)	Dilute 1:10000 in media (DMEM+10%FBS) or (N3 media)

Live Hoechst staining

Expected time: 30 mins

1. Remove media and add Hoechst working solution, diluted using the media of the cells in culture.

2. Incubate cells back in 37°C incubator for 20mins.

3. Wash off the Hoechst solution and replace with appropriate culture media.

4. Image stained nuclei using Keyence microscope or analyze cells by flow in the Attune.
