Culturing Jurkat Cells

Jurkat cells grow in suspension. Many of the techniques parallel culturing adherent cells. The notable differents are described here.

Media

RPMI 1640, 10% FBS .. note:: Optional: 1X NEAA (not used in our lab.)

Culturing

Culture cells in suspension with media. Cells can be cultured in T75 or T182 in the 37C incubator. Rocking in not neccesary for cell growth.

Passaging & Media Changes

Spin down cells from suspension for 4 minutes at 400 G. Resuspend cells in RPMI 1640 (10% FBS). Replace cells into a culture flask at the desired passaging ratio.

Cryopreservation

Resuspended cells after centrifugation can be cryopreserved and thawed as described under cryopreservation and thawing.
