FISH buffer recipes
Probe Hybridization Buffer
- Combine the following components fresh before use

| 30% Formamide | 3 mL | 
| 20X SSC | 2.5 mL | 
| 1M Citric Acid, pH 6 | 90 uL | 
| 10% Tween 20 | 100 uL | 
| 50X Denhardt’s Solution | 200 uL | 
| 50% Dextran Sulfate | 1.5 mL | 
| 20 mg/mL BSA | 50 uL | 
| H20 | Bring volume up to 10 mL | 
Probe Wash Buffer
- Combine the following components 
| Ingredient | Amount | 
|---|---|
| 30% Formamide | 3 mL | 
| 20X SSC | 2.5 mL | 
| 1M Citric Acid, pH 6 | 90 uL | 
| 10% Tween 20 | 100 uL | 
| H20 | Bring volume up to 10 mL | 
- Store at -20 degrees C, expiration in 7 days 
Probe Amplification Buffer
- Combine the following components:

| Na2HPO4 | 70.98 mg | 
| NaCL | 292.2 mg | 
| H20 | Bring volume up to 10 mL | 
- Bring the pH to 6.8 (approximately 50 uL of 2.4 M HCl). Store at room temp. 
5X SSCT
- Combine the following components 
| Ingredient | Amount | 
|---|---|
| 20X SSC | 10 mL | 
| 10% Tween 20 | 400 uL | 
| DEPC H20 | Bring volume up to 40 mL | 
- Store at 4 degrees C, expiration in 1 month.
